Zemprocitinib trial results showed sustained treatment responses through week 76 in adults with moderate-to-severe rheumatoid arthritis, developer Lynk Pharmaceuticals said on October 28, 2026. The Phase III study tested the investigational oral drug in patients who had an inadequate response or intolerance to prior biologic therapies.
Key Takeaways
- The trial met its primary and key secondary efficacy endpoints at week 24, Lynk said.
- Patients receiving continuous zemprocitinib treatment maintained high response rates through week 76.
- Lynk said no new safety signals emerged during long-term treatment.
Zemprocitinib trial results through week 76
The study randomized 430 patients, of whom 429 received at least one dose. During the first 24 weeks, patients received either 12 mg of zemprocitinib twice daily or a placebo. Afterward, they continued or switched to active treatment.
Lynk said zemprocitinib produced statistically significant improvements over placebo at week 24 on the primary ACR20 endpoint and key secondary endpoints. The company reported the following longer-term response rates for patients receiving continuous treatment.
| Item | Details |
|---|---|
| ACR20 response | 93.0% at week 52; 91.8% at week 76 |
| ACR50 response | 79.9% at week 52; 80.5% at week 76 |
| ACR70 response | 52.5% at week 52; 62.4% at week 76 |
Safety and development plans
Lynk said the drug’s safety profile was generally comparable to placebo during the initial 24-week period and remained consistent through week 76, with no new safety signals identified. The company did not provide overall adverse-event rates in the release.
Zemprocitinib is also being developed for atopic dermatitis and ankylosing spondylitis. A new drug application for atopic dermatitis has been accepted for review by China’s National Medical Products Administration, while a Phase III program in ankylosing spondylitis is underway.
Lynk and Simcere Pharmaceutical Group entered a commercialization collaboration in March 2022 for zemprocitinib in rheumatoid arthritis and ankylosing spondylitis in Greater China.
